The encoded protein also plays a role in the regulation of epidermal growth factor receptor turnover and actin remodelling. Additionally, this gene supports metastatic growth in breast cancer and is a valuable biomarker for breast cancer. [provided by RefSeq, Jan 2009]. * **<color #00a2e8>UniProt Summary</color>**: Phosphatidylinositol (PtdIns) phosphatase that specifically hydrolyzes the 5-phosphate of phosphatidylinositol- 3,4,5-trisphosphate (PtdIns(3,4,5)P3) to produce PtdIns(3,4)P2, thereby negatively regulating the PI3K (phosphoinositide 3-kinase) pathways. Plays a central role in regulation of PI3K-dependent insulin signaling, although the precise molecular mechanisms and signaling pathways remain unclear. While overexpression reduces both insulin-stimulated MAP kinase and Akt activation, its absence does not affect insulin signaling or GLUT4 trafficking. Confers resistance to dietary obesity. May act by regulating AKT2, but not AKT1, phosphorylation at the plasma membrane. Part of a signaling pathway that regulates actin cytoskeleton remodeling. Required for the maintenance and dynamic remodeling of actin structures as well as in endocytosis, having a major impact on ligand-induced EGFR internalization and degradation. Participates in regulation of cortical and submembraneous actin by hydrolyzing PtdIns(3,4,5)P3 thereby regulating membrane ruffling (PubMed:21624956). Regulates cell adhesion and cell spreading. Required for HGF-mediated lamellipodium formation, cell scattering and spreading. Acts as a negative regulator of EPHA2 receptor endocytosis by inhibiting via PI3K-dependent Rac1 activation. Acts as a regulator of neuritogenesis by regulating PtdIns(3,4,5)P3 level and is required to form an initial protrusive pattern, and later, maintain proper neurite outgrowth. Acts as a negative regulator of the FC-gamma- RIIA receptor (FCGR2A). Mediates signaling from the FC-gamma-RIIB receptor (FCGR2B), playing a central role in terminating signal transduction from activating immune/hematopoietic cell receptor systems. Involved in EGF signaling pathway. Upon stimulation by EGF, it is recruited by EGFR and dephosphorylates PtdIns(3,4,5)P3. Plays a negative role in regulating the PI3K-PKB pathway, possibly by inhibiting PKB activity. Down-regulates Fc-gamma-R-mediated phagocytosis in macrophages independently of INPP5D/SHIP1. In macrophages, down-regulates NF-kappa-B-dependent gene transcription by regulating macrophage colony-stimulating factor (M-CSF)-induced signaling. May also hydrolyze PtdIns(1,3,4,5)P4, and could thus affect the levels of the higher inositol polyphosphates like InsP6. Involved in endochondral ossification. {ECO:0000269|PubMed:11349134, ECO:0000269|PubMed:11739414, ECO:0000269|PubMed:12235291, ECO:0000269|PubMed:12676785, ECO:0000269|PubMed:12690104, ECO:0000269|PubMed:15668240, ECO:0000269|PubMed:17135240, ECO:0000269|PubMed:21624956, ECO:0000269|PubMed:23273569, ECO:0000269|PubMed:9660833}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |SAM 1| |SAM 2| |Exo endo phos| |SH2| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |ruffle assembly| |phosphatidylinositol dephosphorylation| |ruffle organization| |replacement ossification| |endochondral ossification| |phospholipid dephosphorylation| |SH2 domain binding| |inositol phosphate metabolic process| |endochondral bone morphogenesis| |filopodium| |hydrolase activity| |post-embryonic development| |phosphatidylinositol biosynthetic process| |polyol metabolic process| |bone morphogenesis| |glucose metabolic process| |SH3 domain binding| |hexose metabolic process| |phosphatidylinositol metabolic process| |lamellipodium| |monosaccharide metabolic process| |bone development| |lipid modification| |glycerophospholipid biosynthetic process| |glycerolipid biosynthetic process| |response to insulin| |skeletal system morphogenesis| |actin filament organization| |phospholipid biosynthetic process| |ossification| |actin binding| |glycerophospholipid metabolic process| |dephosphorylation| |alcohol metabolic process| |cytoskeleton| |phospholipid metabolic process| |glycerolipid metabolic process| |response to peptide hormone| |nuclear speck| |plasma membrane bounded cell projection assembly| |organic hydroxy compound metabolic process| |supramolecular fiber organization| |cell projection assembly| |response to peptide| |carbohydrate metabolic process| |actin cytoskeleton organization| |skeletal system development| |organophosphate biosynthetic process| |endocytosis| |actin filament-based process| |lipid biosynthetic process| |cytokine-mediated signaling pathway| |negative regulation of cell population proliferation| |import into cell| |organophosphate metabolic process| |response to hormone| |cell adhesion| |biological adhesion| |cellular lipid metabolic process| |animal organ morphogenesis| |Golgi apparatus| |response to organonitrogen compound| |cellular response to cytokine stimulus| |response to nitrogen compound| |response to cytokine| |cytoskeleton organization| |plasma membrane bounded cell projection organization| |cell projection organization| |lipid metabolic process| |response to endogenous stimulus| |response to oxygen-containing compound| |regulation of cell population proliferation| |negative regulation of gene expression| |small molecule metabolic process| |vesicle-mediated transport| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |S-trityl-L-cysteine 0.5μM R05 exp243|-1.97| |Cabazitaxel 0.001μM R05 exp223|-1.89| |A-395N 10μM R05 exp219|-1.73| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 7514 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 7.04 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='INPPL1 Expression in NALM6 Cells: 7.04'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
