The protein contains both SH2 and SH3 domains which are often found in intracellular kinases. It is thought to play a role in T-cell proliferation and differentiation. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: Tyrosine kinase that plays an essential role in regulation of the adaptive immune response. Regulates the development, function and differentiation of conventional T-cells and nonconventional NKT-cells. When antigen presenting cells (APC) activate T-cell receptor (TCR), a series of phosphorylation lead to the recruitment of ITK to the cell membrane, in the vicinity of the stimulated TCR receptor, where it is phosphorylated by LCK. Phosphorylation leads to ITK autophosphorylation and full activation. Once activated, phosphorylates PLCG1, leading to the activation of this lipase and subsequent cleavage of its substrates. In turn, the endoplasmic reticulum releases calcium in the cytoplasm and the nuclear activator of activated T-cells (NFAT) translocates into the nucleus to perform its transcriptional duty. Phosphorylates 2 essential adapter proteins: the linker for activation of T-cells/LAT protein and LCP2. Then, a large number of signaling molecules such as VAV1 are recruited and ultimately lead to lymphokine production, T-cell proliferation and differentiation (PubMed:12186560, PubMed:12682224, PubMed:21725281). Phosphorylates TBX21 at 'Tyr-530' and mediates its interaction with GATA3 (By similarity). {ECO:0000250|UniProtKB:Q03526, ECO:0000269|PubMed:12186560, ECO:0000269|PubMed:12682224, ECO:0000269|PubMed:21725281}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |SH2| |Pkinase Tyr| |BTK| |SH3 1| |Pkinase| |PH| |SH3 2| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |NK T cell differentiation| |interleukin-4 production| |interferon-gamma production| |activation of phospholipase C activity| |peptidyl-tyrosine autophosphorylation| |positive regulation of phospholipase C activity| |non-membrane spanning protein tyrosine kinase activity| |regulation of phospholipase C activity| |alpha-beta T cell differentiation| |cellular defense response| |positive regulation of phospholipase activity| |alpha-beta T cell activation| |regulation of phospholipase activity| |positive regulation of lipase activity| |regulation of lipase activity| |B cell receptor signaling pathway| |T cell differentiation| |peptidyl-tyrosine phosphorylation| |peptidyl-tyrosine modification| |cytokine production| |cell-cell junction| |Fc-epsilon receptor signaling pathway| |T cell receptor signaling pathway| |protein autophosphorylation| |T cell activation| |lymphocyte differentiation| |Fc receptor signaling pathway| |antigen receptor-mediated signaling pathway| |leukocyte differentiation| |lymphocyte activation| |immune response-activating cell surface receptor signaling pathway| |immune response-regulating cell surface receptor signaling pathway| |hemopoiesis| |immune response-activating signal transduction| |immune response-regulating signaling pathway| |hematopoietic or lymphoid organ development| |adaptive immune response| |activation of immune response| |immune system development| |positive regulation of hydrolase activity| |positive regulation of immune response| |peptidyl-amino acid modification| |leukocyte activation| |protein phosphorylation| |cell activation| |positive regulation of immune system process| |regulation of immune response| |regulation of hydrolase activity| |phosphorylation| |defense response| |positive regulation of catalytic activity| |ATP binding| |regulation of immune system process| |intracellular signal transduction| |positive regulation of molecular function| |immune response| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |GSK-J4 1.5μM R08 exp441|-2.17| |Trametinib 10μM R02 exp83|-1.96| |FBS-Wisent 0.1 R07 exp426|1.95| |Tubastatin-A 2.5μM R07 exp417|2| |Ammonium tetrathiomolybdate 10μM R08 exp448|2.07| |Centrinone 0.5μM R07 exp343|2.28| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 12697 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 3.37 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='ITK Expression in NALM6 Cells: 3.37'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
