The hexameric protein complex formed by MCM proteins is a key component of the pre-replication complex (pre_RC) and may be involved in the formation of replication forks and in the recruitment of other DNA replication related proteins. The MCM complex consisting of this protein and MCM2, 6 and 7 proteins possesses DNA helicase activity, and may act as a DNA unwinding enzyme. The phosphorylation of this protein by CDC2 kinase reduces the DNA helicase activity and chromatin binding of the MCM complex. This gene is mapped to a region on the chromosome 8 head-to-head next to the PRKDC/DNA-PK, a DNA-activated protein kinase involved in the repair of DNA double-strand breaks. Alternatively spliced transcript variants encoding the same protein have been reported. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: Acts as component of the MCM2-7 complex (MCM complex) which is the putative replicative helicase essential for 'once per cell cycle' DNA replication initiation and elongation in eukaryotic cells. The active ATPase sites in the MCM2-7 ring are formed through the interaction surfaces of two neighboring subunits such that a critical structure of a conserved arginine finger motif is provided in trans relative to the ATP-binding site of the Walker A box of the adjacent subunit. The six ATPase active sites, however, are likely to contribute differentially to the complex helicase activity. {ECO:0000269|PubMed:16899510, ECO:0000269|PubMed:9305914}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |AAA 5| |MCM| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |nuclear cell cycle DNA replication initiation| |cell cycle DNA replication initiation| |mitotic DNA replication initiation| |pre-replicative complex assembly involved in nuclear cell cycle DNA replication| |pre-replicative complex assembly involved in cell cycle DNA replication| |pre-replicative complex assembly| |mitotic DNA replication| |double-strand break repair via break-induced replication| |MCM complex| |DNA unwinding involved in DNA replication| |DNA strand elongation involved in DNA replication| |DNA strand elongation| |DNA replication origin binding| |DNA replication initiation| |nuclear DNA replication| |cell cycle DNA replication| |DNA helicase activity| |double-strand break repair via homologous recombination| |recombinational repair| |single-stranded DNA binding| |nuclear chromosome, telomeric region| |DNA duplex unwinding| |DNA geometric change| |G1/S transition of mitotic cell cycle| |DNA-dependent DNA replication| |cell cycle G1/S phase transition| |double-strand break repair| |protein-DNA complex assembly| |DNA replication| |DNA recombination| |protein-DNA complex subunit organization| |mitotic cell cycle phase transition| |cell cycle phase transition| |DNA conformation change| |DNA repair| |mitotic cell cycle process| |mitotic cell cycle| |DNA metabolic process| |cellular response to DNA damage stimulus| |cellular protein-containing complex assembly| |cell cycle process| |chromosome organization| |cell cycle| |ATP binding| |protein-containing complex assembly| |cellular response to stress| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |protein-containing complex subunit organization| |membrane| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |THZ531 0.225μM R08 exp444|-1.8| |CAY10603 0.55μM R08 exp467|-1.78| |Torin1 0.08μM R02 exp82|-1.74| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |HGC6.3|0.615| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 639/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|1/1| |909776.0|1/1| |bile duct|23/28| |blood|24/28| |bone|22/26| |breast|27/33| |central nervous system|53/56| |cervix|3/4| |colorectal|16/17| |esophagus|10/13| |fibroblast|1/1| |gastric|15/16| |kidney|18/21| |liver|16/20| |lung|65/75| |lymphocyte|15/16| |ovary|21/26| |pancreas|23/24| |peripheral nervous system|15/16| |plasma cell|11/15| |prostate|1/1| |skin|21/24| |soft tissue|8/9| |thyroid|2/2| |upper aerodigestive|10/22| |urinary tract|24/29| |uterus|4/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 1127 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 8.77 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='MCM4 Expression in NALM6 Cells: 8.77'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
