The protein consists of a motor domain containing an ATP- and an actin-binding site and a globular tail which interacts with other proteins. This protein maintains the structural integrity of inner ear hair cells and mutations in this gene cause non-syndromic autosomal dominant and recessive hearing loss. Alternative splicing results in multiple transcript variants encoding distinct isoforms. Myosin 6 is a reverse-direction motor protein that moves towards the minus-end of actin filaments. Has slow rate of actin-activated ADP release due to weak ATP binding. Functions in a variety of intracellular processes such as vesicular membrane trafficking and cell migration. Required for the structural integrity of the Golgi apparatus via the p53-dependent pro-survival pathway. Appears to be involved in a very early step of clathrin-mediated endocytosis in polarized epithelial cells. May act as a regulator of F-actin dynamics. May play a role in transporting DAB2 from the plasma membrane to specific cellular targets. Required for structural integrity of inner ear hair cells (By similarity). {ECO:0000250, ECO:0000269|PubMed:10519557, ECO:0000269|PubMed:11447109, ECO:0000269|PubMed:16507995, ECO:0000269|PubMed:16949370}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Myosin head| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |minus-end directed microfilament motor activity| |unconventional myosin complex| |RNA polymerase II, holoenzyme| |clathrin-coated endocytic vesicle| |filamentous actin| |ADP binding| |motor activity| |clathrin-coated vesicle membrane| |clathrin-coated pit| |endocytic vesicle| |microvillus| |apical part of cell| |filopodium| |DNA damage response, signal transduction by p53 class mediator| |actin filament| |ruffle| |ruffle membrane| |signal transduction in response to DNA damage| |actin filament-based movement| |signal transduction by p53 class mediator| |cell cortex| |sensory perception of sound| |sensory perception of mechanical stimulus| |calmodulin binding| |actin filament binding| |nuclear membrane| |cytoplasmic vesicle| |actin binding| |lysosomal membrane| |endocytosis| |actin filament-based process| |import into cell| |perinuclear region of cytoplasm| |cellular response to DNA damage stimulus| |regulation of secretion| |sensory perception| |Golgi apparatus| |intracellular protein transport| |response to drug| |positive regulation of transcription by RNA polymerase II| |nervous system process| |ATP binding| |protein transport| |intracellular transport| |peptide transport| |positive regulation of transcription, DNA-templated| |movement of cell or subcellular component| |amide transport| |cellular protein localization| |cellular macromolecule localization| |establishment of protein localization| |positive regulation of nucleic acid-templated transcription| |positive regulation of RNA biosynthetic process| |intracellular signal transduction| |cellular response to stress| |positive regulation of RNA metabolic process| |establishment of localization in cell| |nitrogen compound transport| |regulation of transport| |positive regulation of nucleobase-containing compound metabolic process| |positive regulation of macromolecule biosynthetic process| |vesicle-mediated transport| |system process| |positive regulation of cellular biosynthetic process| |positive regulation of gene expression| |membrane| |positive regulation of biosynthetic process| </modal> \\ === CRISPR Data === <button type='default' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> No hits were found. </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 15118 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 2.02 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='MYO6 Expression in NALM6 Cells: 2.02'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
