The NELF complex, which acts via an association with the DSIF complex and causes transcriptional pausing, is counteracted by the P-TEFb kinase complex. The NELF complex is involved in HIV-1 latency possibly involving recruitment of PCF11 to paused RNA polymerase II. Binds RNA which may help to stabilize the NELF complex on nucleic acid. In vitro, binds weakly to the HIV-1 TAR RNA which is located in the long terminal repeat (LTR) of HIV-1. May be able to induce chromatin unfolding. {ECO:0000269|PubMed:10199401, ECO:0000269|PubMed:12612062, ECO:0000269|PubMed:23884411, ECO:0000269|PubMed:27282391}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |COBRA1| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |NELF complex| |negative regulation of transcription elongation from RNA polymerase II promoter| |negative regulation of DNA-templated transcription, elongation| |negative regulation of stem cell differentiation| |regulation of transcription elongation from RNA polymerase II promoter| |positive regulation of viral transcription| |regulation of DNA-templated transcription, elongation| |regulation of viral transcription| |transcription elongation from RNA polymerase II promoter| |DNA-templated transcription, elongation| |positive regulation of viral process| |regulation of stem cell differentiation| |stem cell differentiation| |regulation of viral process| |regulation of symbiosis, encompassing mutualism through parasitism| |transcription by RNA polymerase II| |positive regulation of multi-organism process| |cell population proliferation| |transcription, DNA-templated| |nucleic acid-templated transcription| |RNA biosynthetic process| |negative regulation of cell differentiation| |regulation of multi-organism process| |negative regulation of transcription by RNA polymerase II| |negative regulation of developmental process| |nucleobase-containing compound biosynthetic process| |heterocycle biosynthetic process| |aromatic compound biosynthetic process| |negative regulation of transcription, DNA-templated| |negative regulation of nucleic acid-templated transcription| |negative regulation of RNA biosynthetic process| |organic cyclic compound biosynthetic process| |negative regulation of RNA metabolic process| |negative regulation of cellular macromolecule biosynthetic process| |RNA binding| |negative regulation of nucleobase-containing compound metabolic process| |negative regulation of macromolecule biosynthetic process| |negative regulation of cellular biosynthetic process| |negative regulation of biosynthetic process| |cellular nitrogen compound biosynthetic process| |RNA metabolic process| |cellular macromolecule biosynthetic process| |negative regulation of gene expression| |macromolecule biosynthetic process| |regulation of cell differentiation| |gene expression| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |JQ1 0.8μM R08 exp435|-3.24| |PU-H71 1μM R08 exp515|-2.35| |Deferoxamine 11μM R07 exp350|-2.29| |Pimelic-diphenylamide-106 5μM R00 exp28|-2.22| |BVD-523 15μM R02 exp67|-2.18| |BI-D1870 3.15μM R02 exp66|-1.95| |Dinaciclib 0.007μM R07 exp355|-1.77| |Prochlorperazine 5.2μM R02 exp77|1.74| |Deguelin 0.05μM R02 exp69|2.05| |Mubritinib 0.2μM R02 exp65|2.18| |Trimetrexate 0.03μM R08 exp535|2.76| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |NELFA|0.508| |BRD2|0.473| |PDS5B|0.472| |NELFCD|0.421| |IPPK|0.409| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 708/726 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|1/1| |bile duct|27/28| |blood|27/28| |bone|25/25| |breast|31/33| |central nervous system|56/56| |cervix|4/4| |colorectal|17/17| |esophagus|12/13| |fibroblast|1/1| |gastric|14/15| |kidney|20/21| |liver|20/20| |lung|73/75| |lymphocyte|14/14| |ovary|26/26| |pancreas|24/24| |peripheral nervous system|16/16| |plasma cell|15/15| |prostate|1/1| |skin|24/24| |soft tissue|6/7| |thyroid|2/2| |upper aerodigestive|22/22| |urinary tract|28/29| |uterus|5/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 138 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 6.37 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='NELFB Expression in NALM6 Cells: 6.37'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
