Plays a key role in the structural and functional plasticity of neurons (By similarity). Acts as an early-response transcription factor in both excitatory and inhibitory neurons, where it induces distinct but overlapping sets of late-response genes in these two types of neurons, allowing the synapses that form on inhibitory and excitatory neurons to be modified by neuronal activity in a manner specific to their function within a circuit, thereby facilitating appropriate circuit responses to sensory experience (By similarity). In excitatory neurons, activates transcription of BDNF, which in turn controls the number of GABA-releasing synapses that form on excitatory neurons, thereby promoting an increased number of inhibitory synapses on excitatory neurons (By similarity). In inhibitory neurons, regulates a distinct set of target genes that serve to increase excitatory input onto somatostatin neurons, probably resulting in enhanced feedback inhibition within cortical circuits (By similarity). The excitatory and inhibitory balance in neurons affects a number of processes, such as short-term and long-term memory, acquisition of experience, fear memory, response to stress and social behavior (By similarity). Acts as a regulator of dendritic spine development in olfactory bulb granule cells in a sensory-experience-dependent manner by regulating expression of MDM2 (By similarity). Efficient DNA binding requires dimerization with another bHLH protein, such as ARNT, ARNT2 or BMAL1 (PubMed:14701734). Can activate the CME (CNS midline enhancer) element (PubMed:14701734). {ECO:0000250|UniProtKB:Q8BGD7, ECO:0000269|PubMed:14701734}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |PAS 3| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |cellular response to corticosterone stimulus| |cellular response to mineralocorticoid stimulus| |short-term memory| |inhibitory synapse assembly| |inhibitory postsynaptic potential| |response to corticosterone| |regulation of synaptic transmission, GABAergic| |response to mineralocorticoid| |long-term memory| |intraspecies interaction between organisms| |social behavior| |cellular response to glucocorticoid stimulus| |cellular response to corticosteroid stimulus| |excitatory postsynaptic potential| |chemical synaptic transmission, postsynaptic| |multi-organism behavior| |postsynapse| |cellular response to alcohol| |cellular response to ketone| |synapse assembly| |regulation of postsynaptic membrane potential| |memory| |response to glucocorticoid| |learning| |response to corticosteroid| |cellular response to steroid hormone stimulus| |regulation of synaptic plasticity| |response to ketone| |transcription factor complex| |response to alcohol| |learning or memory| |synapse organization| |cognition| |response to steroid hormone| |chemical synaptic transmission| |anterograde trans-synaptic signaling| |cell surface receptor signaling pathway involved in cell-cell signaling| |regulation of membrane potential| |trans-synaptic signaling| |modulation of chemical synaptic transmission| |regulation of trans-synaptic signaling| |DNA-binding transcription activator activity, RNA polymerase II-specific| |synaptic signaling| |protein heterodimerization activity| |RNA polymerase II proximal promoter sequence-specific DNA binding| |cellular response to lipid| |cellular response to organic cyclic compound| |behavior| |cellular response to hormone stimulus| |response to lipid| |response to hormone| |response to organic cyclic compound| |cellular response to oxygen-containing compound| |cell-cell signaling| |positive regulation of transcription by RNA polymerase II| |cellular response to endogenous stimulus| |nervous system process| |response to endogenous stimulus| |positive regulation of transcription, DNA-templated| |response to oxygen-containing compound| |DNA-binding transcription factor activity, RNA polymerase II-specific| |positive regulation of nucleic acid-templated transcription| |positive regulation of RNA biosynthetic process| |positive regulation of RNA metabolic process| |positive regulation of nucleobase-containing compound metabolic process| |positive regulation of macromolecule biosynthetic process| |system process| |positive regulation of cellular biosynthetic process| |positive regulation of gene expression| |positive regulation of biosynthetic process| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |[[:results:exp495|IWR1 50μM R08 exp495]]|-1.89| |[[:results:exp347|Cyclosporin-A 0.8μM R07 exp347]]|-1.71| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 8203 * **<color #00a2e8>Expression level (log2 read counts)</color>**: -4.01 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='NPAS4 Expression in NALM6 Cells: -4.01'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
