May regulate the activity of PPP4C at centrosomal microtubule organizing centers. Its interaction with the SMN complex leads to enhance the temporal localization of snRNPs, suggesting a role of PPP4C in maturation of spliceosomal snRNPs. The PPP4C-PPP4R2-PPP4R3A PP4 complex specifically dephosphorylates H2AFX phosphorylated on 'Ser-140' (gamma-H2AFX) generated during DNA replication and required for DNA double strand break repair. Mediates RPA2 dephosphorylation by recruiting PPP4C to RPA2 in a DNA damage-dependent manner. RPA2 dephosphorylation is required for the efficient RPA2-mediated recruitment of RAD51 to chromatin following double strand breaks, an essential step for DNA repair. {ECO:0000269|PubMed:10769191, ECO:0000269|PubMed:12668731, ECO:0000269|PubMed:18614045, ECO:0000269|PubMed:20154705}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |PPP4R2| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |protein phosphatase 4 complex| |protein phosphatase regulator activity| |regulation of double-strand break repair via homologous recombination| |regulation of double-strand break repair| |protein binding, bridging| |regulation of DNA recombination| |regulation of phosphoprotein phosphatase activity| |regulation of DNA repair| |regulation of protein dephosphorylation| |regulation of phosphatase activity| |regulation of dephosphorylation| |protein dephosphorylation| |regulation of response to DNA damage stimulus| |dephosphorylation| |regulation of DNA metabolic process| |RNA splicing| |mRNA processing| |centrosome| |mRNA metabolic process| |regulation of cellular response to stress| |RNA processing| |regulation of hydrolase activity| |regulation of response to stress| |RNA metabolic process| |regulation of phosphate metabolic process| |regulation of phosphorus metabolic process| |regulation of protein modification process| |gene expression| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Rapamycin 0.001μM R08 exp429|-1.95| |Docetaxel 0.002μM R01 exp45|-1.94| |Adefovir 20μM R07 exp332|-1.94| |Rapamycin 1μM R00 exp29|-1.82| |SGC2096 2.6μM R03 exp153|1.72| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 55/726 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|2/28| |blood|2/28| |bone|5/25| |breast|3/33| |central nervous system|7/56| |cervix|0/4| |colorectal|1/17| |esophagus|0/13| |fibroblast|0/1| |gastric|1/15| |kidney|3/21| |liver|2/20| |lung|3/75| |lymphocyte|0/14| |ovary|3/26| |pancreas|4/24| |peripheral nervous system|0/16| |plasma cell|1/15| |prostate|0/1| |skin|2/24| |soft tissue|0/7| |thyroid|0/2| |upper aerodigestive|1/22| |urinary tract|1/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 2931 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 7.02 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='PPP4R2 Expression in NALM6 Cells: 7.02'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
