This protein contains a RING zinc-finger motif and has been shown to possess E3 ubiquitin ligase activity. Expression of this gene in retrovirally transduced T cell hybridoma significantly inhibits activation-induced IL2 and IL4 cytokine production. Induced expression of this gene was observed in anergic CD4(+) T cells, which suggested a role in the induction of anergic phenotype. Alternatively spliced transcript variants encoding distinct isoforms have been reported. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: E3 ubiquitin-protein ligase that catalyzes 'Lys-48'- and 'Lys-63'-linked polyubiquitin chains formation. Functions as an inhibitor of cytokine gene transcription. Inhibits IL2 and IL4 transcription, thereby playing an important role in the induction of the anergic phenotype, a long-term stable state of T-lymphocyte unresponsiveness to antigenic stimulation associated with the blockade of interleukin production. Ubiquitinates ARPC5 with 'Lys- 48' linkages and COR1A with 'Lys-63' linkages leading to their degradation, down-regulation of these cytosleletal components results in impaired lamellipodium formation and reduced accumulation of F-actin at the immunological synapse. Functions in the patterning of the dorsal ectoderm; sensitizes ectoderm to respond to neural-inducing signals. {ECO:0000269|PubMed:12705856, ECO:0000269|PubMed:22016387}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |zf-C3HC4| |PA| |zf-rbx1| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |positive regulation of protein catabolic process in the vacuole| |regulation of protein catabolic process in the vacuole| |negative regulation of cytokine biosynthetic process| |protein localization to lysosome| |protein localization to vacuole| |regulation of cytokine biosynthetic process| |late endosome| |positive regulation of cellular protein catabolic process| |positive regulation of protein catabolic process| |ubiquitin protein ligase activity| |regulation of cellular protein catabolic process| |negative regulation of cytokine production| |protein deubiquitination| |regulation of protein stability| |protein modification by small protein removal| |positive regulation of cellular catabolic process| |cytoskeleton| |regulation of protein catabolic process| |positive regulation of catabolic process| |ubiquitin-dependent protein catabolic process| |modification-dependent protein catabolic process| |modification-dependent macromolecule catabolic process| |proteolysis involved in cellular protein catabolic process| |cellular protein catabolic process| |protein catabolic process| |protein ubiquitination| |regulation of cytokine production| |perinuclear region of cytoplasm| |protein localization to organelle| |protein modification by small protein conjugation| |regulation of cellular catabolic process| |cellular macromolecule catabolic process| |Golgi apparatus| |protein modification by small protein conjugation or removal| |regulation of catabolic process| |endoplasmic reticulum| |macromolecule catabolic process| |organonitrogen compound catabolic process| |negative regulation of protein metabolic process| |negative regulation of multicellular organismal process| |proteolysis| |negative regulation of macromolecule biosynthetic process| |negative regulation of cellular biosynthetic process| |negative regulation of biosynthetic process| |cellular protein localization| |cellular macromolecule localization| |positive regulation of cellular protein metabolic process| |positive regulation of protein metabolic process| |organic substance catabolic process| |cellular catabolic process| </modal> \\ === CRISPR Data === <button type='default' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> No hits were found. </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/694 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/26| |bone|0/26| |breast|0/30| |central nervous system|0/49| |cervix|0/4| |colorectal|0/17| |esophagus|0/11| |fibroblast|0/1| |gastric|0/14| |kidney|0/18| |liver|0/19| |lung|0/72| |lymphocyte|0/16| |ovary|0/25| |pancreas|0/22| |peripheral nervous system|0/15| |plasma cell|0/12| |prostate|0/1| |skin|0/20| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/28| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 18388 * **<color #00a2e8>Expression level (log2 read counts)</color>**: -7.68 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='RNF128 Expression in NALM6 Cells: -7.68'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
