It is preferentially expressed in the skeletal muscle, and binds to the M-CAT regulatory element found in promoters of muscle-specific genes to direct their gene expression. Alternatively spliced transcripts encoding distinct isoforms, some of which are translated through the use of a non-AUG (UUG) initiation codon, have been described for this gene. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: Transcription factor which plays a key role in the Hippo signaling pathway, a pathway involved in organ size control and tumor suppression by restricting proliferation and promoting apoptosis. The core of this pathway is composed of a kinase cascade wherein MST1/MST2, in complex with its regulatory protein SAV1, phosphorylates and activates LATS1/2 in complex with its regulatory protein MOB1, which in turn phosphorylates and inactivates YAP1 oncoprotein and WWTR1/TAZ. Acts by mediating gene expression of YAP1 and WWTR1/TAZ, thereby regulating cell proliferation, migration and epithelial mesenchymal transition (EMT) induction. Binds specifically and non-cooperatively to the Sph and GT-IIC 'enhansons' (5'-GTGGAATGT-3') and activates transcription. Binds to the M-CAT motif. {ECO:0000269|PubMed:18579750, ECO:0000269|PubMed:19324877}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |TEA| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |hippo signaling| |RNA polymerase II transcription factor binding| |transcription initiation from RNA polymerase II promoter| |transcription factor complex| |DNA-templated transcription, initiation| |transcription regulatory region DNA binding| |muscle organ development| |sequence-specific DNA binding| |muscle structure development| |transcription by RNA polymerase II| |skeletal system development| |transcription, DNA-templated| |nucleic acid-templated transcription| |RNA biosynthetic process| |DNA-binding transcription factor activity| |nucleobase-containing compound biosynthetic process| |heterocycle biosynthetic process| |aromatic compound biosynthetic process| |positive regulation of transcription by RNA polymerase II| |organic cyclic compound biosynthetic process| |positive regulation of transcription, DNA-templated| |DNA-binding transcription factor activity, RNA polymerase II-specific| |cellular nitrogen compound biosynthetic process| |positive regulation of nucleic acid-templated transcription| |positive regulation of RNA biosynthetic process| |RNA metabolic process| |intracellular signal transduction| |cellular macromolecule biosynthetic process| |positive regulation of RNA metabolic process| |macromolecule biosynthetic process| |positive regulation of nucleobase-containing compound metabolic process| |positive regulation of macromolecule biosynthetic process| |positive regulation of cellular biosynthetic process| |positive regulation of gene expression| |gene expression| |positive regulation of biosynthetic process| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Golgicide-A 4μM R03 exp122|-1.75| |FK-506 30μM R07 exp359|1.74| |IL-3 9ng/ml R08 exp493|1.94| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |POLR2J3|0.4| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 2/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|1/33| |central nervous system|0/56| |cervix|0/4| |colorectal|1/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 18218 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 4.67 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='TEAD4 Expression in NALM6 Cells: 4.67'> </modal> Last modified: 2026/01/07 22:37by 127.0.0.1
