Believed to act as a core component of the putative HOPS and CORVET endosomal tethering complexes which are proposed to be involved in the Rab5-to-Rab7 endosome conversion probably implicating MON1A/B, and via binding SNAREs and SNARE complexes to mediate tethering and docking events during SNARE-mediated membrane fusion. The HOPS complex is proposed to be recruited to Rab7 on the late endosomal membrane and to regulate late endocytic, phagocytic and autophagic traffic towards lysosomes. The CORVET complex is proposed to function as a Rab5 effector to mediate early endosome fusion probably in specific endosome subpopulations (PubMed:11382755, PubMed:23351085, PubMed:24554770, PubMed:25783203). Required for fusion of endosomes and autophagosomes with lysosomes (PubMed:25783203). Involved in dendrite development of Pukinje cells (By similarity). {ECO:0000250|UniProtKB:Q8R307, ECO:0000269|PubMed:25783203, ECO:0000305|PubMed:11382755, ECO:0000305|PubMed:23351085, ECO:0000305|PubMed:25783203}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Clathrin| |Pep3 Vps18| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |CORVET complex| |AP-3 adaptor complex| |regulation of SNARE complex assembly| |HOPS complex| |regulation of vesicle fusion| |vesicle docking involved in exocytosis| |endosome to lysosome transport| |syntaxin binding| |clathrin-coated vesicle| |lytic vacuole organization| |lysosome organization| |vesicle docking| |exocytic process| |autophagosome| |endosome organization| |regulation of synaptic vesicle exocytosis| |protein binding, bridging| |actin filament| |presynapse| |viral entry into host cell| |entry into host| |entry into host cell| |regulation of neurotransmitter secretion| |lysosomal transport| |regulation of synaptic vesicle cycle| |late endosome membrane| |late endosome| |vacuole organization| |vacuolar transport| |regulation of neurotransmitter transport| |interaction with host| |regulation of regulated secretory pathway| |organelle localization by membrane tethering| |membrane docking| |viral life cycle| |regulation of exocytosis| |lysosome| |early endosome| |process utilizing autophagic mechanism| |autophagy| |actin binding| |endosome| |lysosomal membrane| |vesicle organization| |glutamatergic synapse| |regulation of neurotransmitter levels| |endomembrane system organization| |modulation of chemical synaptic transmission| |regulation of trans-synaptic signaling| |regulation of protein complex assembly| |regulation of vesicle-mediated transport| |organelle localization| |viral process| |regulation of secretion by cell| |symbiotic process| |exocytosis| |regulation of secretion| |interspecies interaction between organisms| |regulation of cellular localization| |regulation of cellular component biogenesis| |intracellular protein transport| |secretion by cell| |export from cell| |secretion| |regulation of organelle organization| |protein transport| |intracellular transport| |peptide transport| |amide transport| |cellular protein localization| |cellular macromolecule localization| |establishment of protein localization| |cellular catabolic process| |establishment of localization in cell| |nitrogen compound transport| |regulation of transport| |vesicle-mediated transport| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Clomiphene 4.4μM R02 exp68|-2.23| |Benzoate 10000μM R08 exp455|-1.85| |Q15 1 to 2μM on day4 R04 exp164|-1.79| |B02 10μM R08 exp453|-1.73| |L-74142 5μM R03 exp131|1.74| |AICAR 240μM R05 exp211|1.83| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 377/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|13/28| |blood|19/28| |bone|8/26| |breast|20/33| |central nervous system|23/56| |cervix|2/4| |colorectal|9/17| |esophagus|4/13| |fibroblast|0/1| |gastric|7/16| |kidney|11/21| |liver|11/20| |lung|44/75| |lymphocyte|12/16| |ovary|11/26| |pancreas|11/24| |peripheral nervous system|5/16| |plasma cell|11/15| |prostate|0/1| |skin|6/24| |soft tissue|6/9| |thyroid|0/2| |upper aerodigestive|14/22| |urinary tract|16/29| |uterus|4/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 1879 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 5.92 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='VPS18 Expression in NALM6 Cells: 5.92'> </modal> Last modified: 2026/01/07 22:37by 127.0.0.1
