NER is responsible for repair of UV radiation-induced photoproducts and DNA adducts induced by chemical carcinogens and chemotherapeutic drugs. The encoded protein interacts with DNA and several NER proteins, acting as a scaffold to assemble the NER incision complex at sites of DNA damage. Mutations in this gene cause Xeroderma pigmentosum complementation group A (XP-A), an autosomal recessive skin disorder featuring hypersensitivity to sunlight and increased risk for skin cancer. [provided by RefSeq, Aug 2017]. * **<color #00a2e8>UniProt Summary</color>**: Involved in DNA excision repair. Initiates repair by binding to damaged sites with various affinities, depending on the photoproduct and the transcriptional state of the region. Required for UV-induced CHEK1 phosphorylation and the recruitment of CEP164 to cyclobutane pyrimidine dimmers (CPD), sites of DNA damage after UV irradiation. {ECO:0000269|PubMed:19197159}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |XPA C| |XPA N| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |nucleotide-excision repair involved in interstrand cross-link repair| |nucleotide-excision repair factor 1 complex| |UV protection| |UV-damage excision repair| |DNA replication factor A complex| |nucleotide-excision repair, preincision complex stabilization| |nucleotide-excision repair, DNA duplex unwinding| |nucleotide-excision repair, DNA incision, 3-to lesion| |nucleotide-excision repair, DNA damage recognition| |response to auditory stimulus| |global genome nucleotide-excision repair| |nucleotide-excision repair, preincision complex assembly| |nucleotide-excision repair, DNA incision, 5-to lesion| |base-excision repair| |nucleotide-excision repair, DNA incision| |interstrand cross-link repair| |damaged DNA binding| |intercellular bridge| |intrinsic apoptotic signaling pathway in response to DNA damage| |transcription-coupled nucleotide-excision repair| |multicellular organism growth| |cellular response to UV| |nucleotide-excision repair| |DNA duplex unwinding| |cellular response to light stimulus| |DNA geometric change| |response to UV| |intrinsic apoptotic signaling pathway| |protein localization to nucleus| |cellular response to radiation| |protein-DNA complex assembly| |response to mechanical stimulus| |protein-DNA complex subunit organization| |protein domain specific binding| |positive regulation of protein complex assembly| |apoptotic signaling pathway| |DNA conformation change| |nucleic acid phosphodiester bond hydrolysis| |response to light stimulus| |cellular response to environmental stimulus| |cellular response to abiotic stimulus| |regulation of autophagy| |response to oxidative stress| |developmental growth| |growth| |response to radiation| |regulation of protein complex assembly| |response to toxic substance| |DNA repair| |positive regulation of cellular component biogenesis| |protein localization to organelle| |DNA metabolic process| |cellular response to DNA damage stimulus| |regulation of cellular catabolic process| |cellular protein-containing complex assembly| |protein homodimerization activity| |apoptotic process| |regulation of cellular component biogenesis| |regulation of catabolic process| |programmed cell death| |chromosome organization| |cell death| |response to abiotic stimulus| |positive regulation of cellular component organization| |protein-containing complex assembly| |cellular protein localization| |cellular macromolecule localization| |intracellular signal transduction| |cellular response to stress| |protein-containing complex subunit organization| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |[[:results:exp423|Zebularine 20μM R07 exp423]]|-1.72| |[[:results:exp462|Cadmium 60μM R08 exp462]]|-1.71| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 1/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|1/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 15877 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 4.11 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='XPA Expression in NALM6 Cells: 4.11'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2026/01/07 22:37by 127.0.0.1
