It is the penultimate enzyme in melatonin synthesis and controls the night/day rhythm in melatonin production in the vertebrate pineal gland. Melatonin is essential for the function of the circadian clock that influences activity and sleep. This enzyme is regulated by cAMP-dependent phosphorylation that promotes its interaction with 14-3-3 proteins and thus protects the enzyme against proteasomal degradation. This gene may contribute to numerous genetic diseases such as delayed sleep phase syndrome. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2009]. * **<color #00a2e8>UniProt Summary</color>**: Controls the night/day rhythm of melatonin production in the pineal gland. Catalyzes the N-acetylation of serotonin into N- acetylserotonin, the penultimate step in the synthesis of melatonin. {ECO:0000269|PubMed:11313340, ECO:0000305}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Acetyltransf 1| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |aralkylamine N-acetyltransferase activity| |melatonin metabolic process| |melatonin biosynthetic process| |arylamine N-acetyltransferase activity| |indole-containing compound biosynthetic process| |indolalkylamine biosynthetic process| |N-terminal protein amino acid acetylation| |indolalkylamine metabolic process| |response to corticosterone| |cellular biogenic amine biosynthetic process| |response to prostaglandin E| |amine biosynthetic process| |indole-containing compound metabolic process| |photoperiodism| |N-terminal protein amino acid modification| |14-3-3 protein binding| |response to prostaglandin| |response to mineralocorticoid| |response to copper ion| |cellular response to cAMP| |response to zinc ion| |cellular biogenic amine metabolic process| |cellular amine metabolic process| |hormone biosynthetic process| |amine metabolic process| |response to fatty acid| |response to cAMP| |cellular hormone metabolic process| |response to organophosphorus| |circadian rhythm| |protein acetylation| |response to glucocorticoid| |response to calcium ion| |response to purine-containing compound| |response to corticosteroid| |protein acylation| |response to ketone| |hormone metabolic process| |protein maturation| |response to insulin| |response to alcohol| |rhythmic process| |response to light stimulus| |response to steroid hormone| |response to acid chemical| |response to metal ion| |response to peptide hormone| |response to radiation| |response to peptide| |drug metabolic process| |amide biosynthetic process| |regulation of hormone levels| |response to inorganic substance| |cellular response to organic cyclic compound| |cellular response to organonitrogen compound| |cellular response to nitrogen compound| |perinuclear region of cytoplasm| |cellular amide metabolic process| |response to lipid| |response to hormone| |response to organic cyclic compound| |response to organonitrogen compound| |cellular response to oxygen-containing compound| |response to nitrogen compound| |response to cytokine| |response to abiotic stimulus| |heterocycle biosynthetic process| |aromatic compound biosynthetic process| |cellular response to endogenous stimulus| |organic cyclic compound biosynthetic process| |organonitrogen compound biosynthetic process| |response to endogenous stimulus| |response to oxygen-containing compound| |cellular nitrogen compound biosynthetic process| |gene expression| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |[[:results:exp212|Phenformin 20μM R05 exp212]]|-1.8| |[[:results:exp115|A-366 10μM R03 exp115]]|-1.75| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 11714 * **<color #00a2e8>Expression level (log2 read counts)</color>**: -2.21 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='AANAT Expression in NALM6 Cells: -2.21'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
