It localizes in mitochondrion and plays an important role in alpha-(+)-lipoic acid synthesis. It may also function in the sulfur insertion chemistry in lipoate biosynthesis. Alternative splicing occurs at this locus and two transcript variants encoding distinct isoforms have been identified. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: Catalyzes the radical-mediated insertion of two sulfur atoms into the C-6 and C-8 positions of the octanoyl moiety bound to the lipoyl domains of lipoate-dependent enzymes, thereby converting the octanoylated domains into lipoylated derivatives. {ECO:0000255|HAMAP-Rule:MF_03123}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Radical SAM| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |lipoate biosynthetic process| |lipoate synthase activity| |lipoate metabolic process| |4 iron, 4 sulfur cluster binding| |neural tube closure| |tube closure| |primary neural tube formation| |neural tube formation| |fatty acid biosynthetic process| |embryonic epithelial tube formation| |epithelial tube formation| |coenzyme biosynthetic process| |morphogenesis of embryonic epithelium| |tube formation| |neural tube development| |sulfur compound biosynthetic process| |monocarboxylic acid biosynthetic process| |cofactor biosynthetic process| |coenzyme metabolic process| |carboxylic acid biosynthetic process| |organic acid biosynthetic process| |epithelial tube morphogenesis| |response to lipopolysaccharide| |fatty acid metabolic process| |response to molecule of bacterial origin| |sulfur compound metabolic process| |mitochondrial matrix| |response to oxidative stress| |morphogenesis of an epithelium| |cofactor metabolic process| |inflammatory response| |monocarboxylic acid metabolic process| |tissue morphogenesis| |embryonic morphogenesis| |lipid biosynthetic process| |small molecule biosynthetic process| |chordate embryonic development| |embryo development ending in birth or egg hatching| |tube morphogenesis| |response to bacterium| |tube development| |response to lipid| |anatomical structure formation involved in morphogenesis| |carboxylic acid metabolic process| |cellular lipid metabolic process| |embryo development| |oxoacid metabolic process| |organic acid metabolic process| |epithelium development| |heterocycle biosynthetic process| |lipid metabolic process| |mitochondrion| |response to other organism| |organic cyclic compound biosynthetic process| |response to external biotic stimulus| |response to biotic stimulus| |defense response| |response to oxygen-containing compound| |small molecule metabolic process| |tissue development| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Nifuroxazide 5μM R03 exp102|-3| |QNZ 0.01μM R05 exp241|-2.6| |HMS-I1 1μM R01 exp46|-2.1| |CCCP 0.1μM R00 exp10|-1.89| |QNZ 0.01μM R08 exp439|-1.87| |Rotenone 0.07μM R08 exp431|-1.86| |Ammonium tetrathiomolybdate 10μM R08 exp448|1.81| |Cannabidiol 13μM R08 exp465|1.92| |Calcium Ionophore 0.4μM R07 exp342|2.21| |AICAR 240μM R03 exp116|2.25| |AICAR 240μM R05 exp211|3.38| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |LIPT1|0.62| |DLD|0.576| |FDX1|0.525| |LIPT2|0.524| |SLC25A19|0.466| |ISCA2|0.446| |GCSH|0.425| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 11/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|1/28| |bone|0/26| |breast|4/33| |central nervous system|0/56| |cervix|1/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|2/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 419 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 5.27 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='LIAS Expression in NALM6 Cells: 5.27'> </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
