Members in this family, classified into six isotypes that are tissue- and organ-specific, hydrolyze phosphatidylinositol 4,5-bisphosphate just before the phosphate group to yield diacylglycerol and inositol 1,4,5-trisphosphate. This protein localizes to the acrosome in spermatozoa and elicits Ca(2+) oscillations and egg activation during fertilization that leads to early embryonic development. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Aug 2012]. * **<color #00a2e8>UniProt Summary</color>**: The production of the second messenger molecules diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3) is mediated by activated phosphatidylinositol-specific phospholipase C enzymes. In vitro, hydrolyzes PtdIns(4,5)P2 in a Ca(2+)- dependent manner. Triggers intracellular Ca(2+) oscillations in oocytes solely during M phase and is involved in inducing oocyte activation and initiating embryonic development up to the blastocyst stage. Is therefore a strong candidate for the egg- activating soluble sperm factor that is transferred from the sperm into the egg cytoplasm following gamete membrane fusion. May exert an inhibitory effect on phospholipase-C-coupled processes that depend on calcium ions and protein kinase C, including CFTR trafficking and function. {ECO:0000250|UniProtKB:Q8K4D7, ECO:0000269|PubMed:12416999, ECO:0000269|PubMed:14697805, ECO:0000269|PubMed:15579586, ECO:0000269|PubMed:26721930, ECO:0000305}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |C2| |PI-PLC-X| |PI-PLC-Y| |efhand like| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |positive regulation of cytosolic calcium ion concentration involved in egg activation| |egg activation| |pronucleus| |sperm head| |inositol trisphosphate biosynthetic process| |phosphatidylinositol-5-phosphate binding| |inositol trisphosphate metabolic process| |phosphatidylinositol phospholipase C activity| |inositol phosphate biosynthetic process| |phosphatidylinositol-3-phosphate binding| |inositol phosphate metabolic process| |polyol biosynthetic process| |phosphatidylinositol-4,5-bisphosphate binding| |polyol metabolic process| |alcohol biosynthetic process| |single fertilization| |fertilization| |organic hydroxy compound biosynthetic process| |calcium ion transport| |positive regulation of cytosolic calcium ion concentration| |divalent metal ion transport| |lipid catabolic process| |divalent inorganic cation transport| |alcohol metabolic process| |regulation of cytosolic calcium ion concentration| |cellular calcium ion homeostasis| |calcium ion homeostasis| |organic hydroxy compound metabolic process| |cellular divalent inorganic cation homeostasis| |divalent inorganic cation homeostasis| |organophosphate biosynthetic process| |cellular metal ion homeostasis| |small molecule biosynthetic process| |metal ion homeostasis| |cellular cation homeostasis| |metal ion transport| |cellular ion homeostasis| |perinuclear region of cytoplasm| |cation homeostasis| |inorganic ion homeostasis| |calcium ion binding| |cellular chemical homeostasis| |ion homeostasis| |cation transport| |sexual reproduction| |nucleolus| |organophosphate metabolic process| |cellular homeostasis| |multi-organism reproductive process| |cell activation| |chemical homeostasis| |lipid metabolic process| |ion transport| |reproductive process| |reproduction| |homeostatic process| |intracellular signal transduction| |small molecule metabolic process| |organic substance catabolic process| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Trimetrexate 0.03μM R08 exp535|-1.82| |GSK626616 14μM R08 exp485|-1.7| |SGC0649 7μM R03 exp150|1.74| |2-Methoxyestradiol 0.2μM R01 exp40|1.8| |Vinblastine 0.002μM R01 exp60|2.17| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 1/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|1/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 13533 * **<color #00a2e8>Expression level (log2 read counts)</color>**: -1.92 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='PLCZ1 Expression in NALM6 Cells: -1.92'> </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
