The encoded protein contains a leucine zipper domain, which is essential for its dimerization with renin. The gene product can catalyze the interconversion of N-acetylglucosamine to N-acetylmannosamine, indicating that it is a GlcNAc 2-epimerase. Transcript variants utilizing alternative promoters have been described in the literature. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: Catalyzes the interconversion of N-acetylglucosamine to N-acetylmannosamine. Binds to renin forming a protein complex called high molecular weight (HMW) renin and inhibits renin activity. Involved in the N-glycolylneuraminic acid (Neu5Gc) degradation pathway: although human is not able to catalyze formation of Neu5Gc due to the inactive CMAHP enzyme, Neu5Gc is present in food and must be degraded. {ECO:0000269|PubMed:9990133}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |GlcNAc 2-epim| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |N-acylglucosamine 2-epimerase activity| |mannosamine metabolic process| |N-acetylmannosamine metabolic process| |N-acetylneuraminate catabolic process| |UDP-N-acetylglucosamine biosynthetic process| |N-acetylneuraminate metabolic process| |amino sugar catabolic process| |amino sugar biosynthetic process| |peptidase inhibitor activity| |UDP-N-acetylglucosamine metabolic process| |N-acetylglucosamine metabolic process| |nucleotide-sugar biosynthetic process| |glucosamine-containing compound metabolic process| |endopeptidase inhibitor activity| |nucleotide-sugar metabolic process| |amino sugar metabolic process| |antibiotic catabolic process| |antibiotic metabolic process| |drug catabolic process| |regulation of blood pressure| |carbohydrate derivative catabolic process| |negative regulation of endopeptidase activity| |negative regulation of peptidase activity| |carboxylic acid catabolic process| |organic acid catabolic process| |negative regulation of proteolysis| |blood circulation| |circulatory system process| |regulation of endopeptidase activity| |small molecule catabolic process| |regulation of peptidase activity| |negative regulation of hydrolase activity| |drug metabolic process| |nucleobase-containing small molecule metabolic process| |carbohydrate derivative biosynthetic process| |regulation of proteolysis| |negative regulation of catalytic activity| |protein homodimerization activity| |carboxylic acid metabolic process| |oxoacid metabolic process| |organic acid metabolic process| |carbohydrate derivative metabolic process| |negative regulation of cellular protein metabolic process| |nucleobase-containing compound biosynthetic process| |negative regulation of protein metabolic process| |negative regulation of molecular function| |heterocycle biosynthetic process| |aromatic compound biosynthetic process| |regulation of hydrolase activity| |organic cyclic compound biosynthetic process| |ATP binding| |cellular nitrogen compound biosynthetic process| |small molecule metabolic process| |organic substance catabolic process| |cellular catabolic process| |system process| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |[[:results:exp133|MKC9989 10μM R03 exp133]]|-1.93| |[[:results:exp42|BI-6727 0.001μM R01 exp42]]|2.1| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/694 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/26| |bone|0/26| |breast|0/30| |central nervous system|0/49| |cervix|0/4| |colorectal|0/17| |esophagus|0/11| |fibroblast|0/1| |gastric|0/14| |kidney|0/18| |liver|0/19| |lung|0/72| |lymphocyte|0/16| |ovary|0/25| |pancreas|0/22| |peripheral nervous system|0/15| |plasma cell|0/12| |prostate|0/1| |skin|0/20| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/28| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 5921 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 2.62 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='RENBP Expression in NALM6 Cells: 2.62'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2025/12/10 20:19by 127.0.0.1
