This protein contains a conserved HMG domain that is involved in DNA binding and nuclear localization, and a conserved C-terminus. Studies suggest that the N-terminal region of this protein interacts with Atxn1 (GeneID:6310), to form a transcription repressor complex, and in vitro studies suggest that polyglutamine-expansion of ATXN1 may alter the repressor activity of this complex. Mutations in this gene have been associated with olidogdendrogliomas (PMID:21817013). In addition, translocation events resulting in gene fusions of this gene with both DUX4 (GeneID:100288687) and FOXO4 (GeneID:4303) have been associated with round cell sarcomas. There are multiple pseudogenes of this gene found on chromosomes 1, 4, 6, 7, 16, 20, and the Y chromosome. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Feb 2015]. * **<color #00a2e8>UniProt Summary</color>**: Transcriptional repressor which plays a role in development of the central nervous system (CNS). In concert with ATXN1 and ATXN1L, involved in brain development. {ECO:0000250|UniProtKB:Q924A2}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |HMG box| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |lung alveolus development| |intraspecies interaction between organisms| |social behavior| |multi-organism behavior| |memory| |learning| |lung development| |respiratory tube development| |respiratory system development| |learning or memory| |cognition| |chromatin binding| |behavior| |protein-containing complex| |intracellular membrane-bounded organelle| |brain development| |head development| |tube development| |negative regulation of transcription by RNA polymerase II| |central nervous system development| |negative regulation of transcription, DNA-templated| |negative regulation of nucleic acid-templated transcription| |negative regulation of RNA biosynthetic process| |negative regulation of RNA metabolic process| |nervous system process| |negative regulation of cellular macromolecule biosynthetic process| |negative regulation of nucleobase-containing compound metabolic process| |DNA binding| |negative regulation of macromolecule biosynthetic process| |negative regulation of cellular biosynthetic process| |negative regulation of biosynthetic process| |DNA-binding transcription factor activity, RNA polymerase II-specific| |negative regulation of gene expression| |system process| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Bortezomib 0.005μM R00 exp6|-2.29| |Phenformin 20μM R05 exp212|-2.28| |GW501516 25μM R06 exp285|-2.06| |S-trityl-L-cysteine 0.5μM R05 exp243|-2.05| |TRAIL 5ng/ml R00 exp35|-2.03| |Rapamycin 10μM R00 exp30|-1.99| |Campthothecin 0.001μM R06 exp270|-1.96| |HMS-I2 1μM R06 exp286|-1.94| |Lead acetate 2000μM R08 exp498 no dilution day6|-1.94| |Citral 75μM R06 exp275|-1.82| |All-trans-Retinoic-Acid 40μM R07 exp334|-1.77| |GSK626616 14μM R08 exp485|-1.77| |Rapamycin 1μM R00 exp29|-1.72| |Vincristine 0.0005μM R04 exp190|-1.71| |Ethambutol 25μM R08 exp481|-1.7| |Palbociclib 1μM R07 exp382|1.71| |Ibrutinib 1μM R07 exp372|1.77| |Ribavirin 10μM R01 exp52|1.84| |BVD-523 15μM R02 exp67|1.91| |Beta-Estradiol 0.55μM R07 exp425|2.06| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |RRM1|0.421| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 5834 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 6.45 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='CIC Expression in NALM6 Cells: 6.45'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
