The esterification of cholesterol is required for cholesterol transport. Mutations in this gene have been found to cause fish-eye disease as well as LCAT deficiency. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: Central enzyme in the extracellular metabolism of plasma lipoproteins. Synthesized mainly in the liver and secreted into plasma where it converts cholesterol and phosphatidylcholines (lecithins) to cholesteryl esters and lysophosphatidylcholines on the surface of high and low density lipoproteins (HDLs and LDLs) (PubMed:10329423, PubMed:19065001, PubMed:26195816). The cholesterol ester is then transported back to the liver. Has a preference for plasma 16:0-18:2 or 18:O-18:2 phosphatidylcholines (PubMed:8820107). Also produced in the brain by primary astrocytes, and esterifies free cholesterol on nascent APOE- containing lipoproteins secreted from glia and influences cerebral spinal fluid (CSF) APOE- and APOA1 levels. Together with APOE and the cholesterol transporter ABCA1, plays a key role in the maturation of glial-derived, nascent lipoproteins. Required for remodeling high-density lipoprotein particles into their spherical forms (PubMed:10722751). {ECO:0000269|PubMed:10329423, ECO:0000269|PubMed:10722751, ECO:0000269|PubMed:14636062, ECO:0000269|PubMed:19065001, ECO:0000269|PubMed:26195816, ECO:0000269|PubMed:8820107}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Abhydrolase 6| |LACT| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |phosphatidylcholine-sterol O-acyltransferase activity| |regulation of high-density lipoprotein particle assembly| |sterol esterification| |steroid esterification| |cholesterol esterification| |apolipoprotein A-I binding| |very-low-density lipoprotein particle remodeling| |triglyceride-rich lipoprotein particle remodeling| |reverse cholesterol transport| |high-density lipoprotein particle remodeling| |high-density lipoprotein particle| |protein-lipid complex remodeling| |plasma lipoprotein particle remodeling| |protein-containing complex remodeling| |phosphatidylcholine biosynthetic process| |plasma lipoprotein particle organization| |protein-lipid complex subunit organization| |cholesterol transport| |sterol transport| |regulation of plasma lipoprotein particle levels| |phosphatidylcholine metabolic process| |cholesterol homeostasis| |sterol homeostasis| |lipoprotein biosynthetic process| |cholesterol metabolic process| |lipoprotein metabolic process| |secondary alcohol metabolic process| |lipid homeostasis| |sterol metabolic process| |organic hydroxy compound transport| |ammonium ion metabolic process| |lipid modification| |glycerophospholipid biosynthetic process| |glycerolipid biosynthetic process| |phospholipid biosynthetic process| |steroid metabolic process| |lipid transport| |glycerophospholipid metabolic process| |alcohol metabolic process| |lipid localization| |phospholipid metabolic process| |glycerolipid metabolic process| |extracellular structure organization| |regulation of protein complex assembly| |organic hydroxy compound metabolic process| |organophosphate biosynthetic process| |lipid biosynthetic process| |organophosphate metabolic process| |cellular lipid metabolic process| |regulation of cellular component biogenesis| |chemical homeostasis| |lipid metabolic process| |organonitrogen compound biosynthetic process| |extracellular space| |homeostatic process| |cellular macromolecule biosynthetic process| |small molecule metabolic process| |macromolecule biosynthetic process| |protein-containing complex subunit organization| |extracellular region| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |[[:results:exp279|D-Fructose 10000μM R06 exp279]]|-1.81| |[[:results:exp51|Nifuroxazide 1μM R01 exp51]]|2.15| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 9393 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 0.9 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='LCAT Expression in NALM6 Cells: 0.9'> {{:chemogenomics:nalm6 dist.png?nolink |}} </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
