The members of this family encode proteins with 50 to 80% sequence identity to each other. The promoters and first exons of the MAGEA genes show considerable variability, suggesting that the existence of this gene family enables the same function to be expressed under different transcriptional controls. The MAGEA genes are clustered at chromosomal location Xq28. They have been implicated in some hereditary disorders, such as dyskeratosis congenita. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: Proposed to enhance ubiquitin ligase activity of RING- type zinc finger-containing E3 ubiquitin-protein ligases. May enhance ubiquitin ligase activity of TRIM28 and stimulate p53/TP53 ubiquitination by TRIM28. Proposed to act through recruitment and/or stabilization of the Ubl-conjugating enzyme (E2) at the E3:substrate complex. May play a role in embryonal development and tumor transformation or aspects of tumor progression. In vitro promotes cell viability in melanoma cell lines. Antigen recognized on a melanoma by autologous cytolytic T-lymphocytes. {ECO:0000269|PubMed:20864041}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |MAGE N| |MAGE| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |caspase binding| |negative regulation of endoplasmic reticulum stress-induced intrinsic apoptotic signaling pathway| |regulation of endoplasmic reticulum stress-induced intrinsic apoptotic signaling pathway| |negative regulation of protein maturation| |negative regulation of protein processing| |negative regulation of response to endoplasmic reticulum stress| |regulation of protein processing| |regulation of protein maturation| |negative regulation of cysteine-type endopeptidase activity involved in apoptotic process| |regulation of response to endoplasmic reticulum stress| |negative regulation of cysteine-type endopeptidase activity| |negative regulation of intrinsic apoptotic signaling pathway| |regulation of intrinsic apoptotic signaling pathway| |regulation of cysteine-type endopeptidase activity involved in apoptotic process| |negative regulation of apoptotic signaling pathway| |regulation of cysteine-type endopeptidase activity| |negative regulation of endopeptidase activity| |negative regulation of peptidase activity| |negative regulation of proteolysis| |regulation of apoptotic signaling pathway| |regulation of endopeptidase activity| |regulation of peptidase activity| |negative regulation of hydrolase activity| |negative regulation of intracellular signal transduction| |regulation of proteolysis| |regulation of cellular response to stress| |negative regulation of catalytic activity| |negative regulation of apoptotic process| |negative regulation of programmed cell death| |negative regulation of cell death| |endoplasmic reticulum| |negative regulation of cellular protein metabolic process| |negative regulation of protein metabolic process| |negative regulation of molecular function| |negative regulation of signal transduction| |regulation of hydrolase activity| |negative regulation of cell communication| |negative regulation of signaling| |regulation of response to stress| |regulation of apoptotic process| |regulation of programmed cell death| |negative regulation of response to stimulus| |regulation of cell death| |negative regulation of gene expression| |regulation of intracellular signal transduction| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Suberoylanilide-Hydroxamic-Acid 0.02μM R01 exp53|-1.87| |BI-2536 0.001μM R01 exp41|-1.78| |Benzoate 10000μM R08 exp455|1.73| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |RRM1|0.477| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/694 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/26| |bone|0/26| |breast|0/30| |central nervous system|0/49| |cervix|0/4| |colorectal|0/17| |esophagus|0/11| |fibroblast|0/1| |gastric|0/14| |kidney|0/18| |liver|0/19| |lung|0/72| |lymphocyte|0/16| |ovary|0/25| |pancreas|0/22| |peripheral nervous system|0/15| |plasma cell|0/12| |prostate|0/1| |skin|0/20| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/28| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 18627 * **<color #00a2e8>Expression level (log2 read counts)</color>**: -7.68 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='MAGEA3 Expression in NALM6 Cells: -7.68'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
