Undergoes restructuring and subcellular redistribution in response to changes in intracellular NADPH/NADP(+) levels. At low NADPH concentrations the protein is found mainly as a monomer, and binds argininosuccinate synthase (ASS1), the enzyme involved in nitric oxide synthesis. Association with ASS1 impairs its activity and reduces the production of nitric oxide, which subsecuently prevents apoptosis. Under normal NADPH concentrations, the protein is found as a dimer and hides the binding site for ASS1. The homodimer binds one molecule of NADPH. Has higher affinity for NADPH than for NADP(+). Binding to NADPH is necessary to form a stable dimer. {ECO:0000269|PubMed:17496144, ECO:0000269|PubMed:18263583, ECO:0000269|PubMed:19254724}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |NmrA| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |nitrogen catabolite repression of transcription| |catabolite repression| |nitrogen catabolite repression of transcription from RNA polymerase II promoter| |nitrogen catabolite regulation of transcription from RNA polymerase II promoter| |nitrogen catabolite regulation of transcription| |regulation of nitrogen utilization| |urea cycle| |urea metabolic process| |nitrogen cycle metabolic process| |cellular response to nutrient| |response to nutrient| |cellular response to nutrient levels| |cellular response to extracellular stimulus| |cellular response to external stimulus| |response to nutrient levels| |amide biosynthetic process| |response to extracellular stimulus| |cellular response to organonitrogen compound| |cellular response to nitrogen compound| |perinuclear region of cytoplasm| |cellular amide metabolic process| |response to organonitrogen compound| |identical protein binding| |response to nitrogen compound| |negative regulation of transcription, DNA-templated| |cellular response to endogenous stimulus| |negative regulation of nucleic acid-templated transcription| |negative regulation of RNA biosynthetic process| |negative regulation of RNA metabolic process| |negative regulation of cellular macromolecule biosynthetic process| |organonitrogen compound biosynthetic process| |negative regulation of nucleobase-containing compound metabolic process| |negative regulation of macromolecule biosynthetic process| |response to endogenous stimulus| |negative regulation of cellular biosynthetic process| |negative regulation of biosynthetic process| |cellular nitrogen compound biosynthetic process| |negative regulation of gene expression| |small molecule metabolic process| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Vinblastine 0.2μM R03 exp108|-2.56| |GSK-J4 1μM R07 exp362|-1.75| |4-Nitroquinoline-1-oxide 0.5μM R08 exp446|4.9| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 6205 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 5.02 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='NMRAL1 Expression in NALM6 Cells: 5.02'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
