The active enzyme is a homotetramer arranged in a tetrahedron which is located exclusively in the mitochondrial matrix. Pyruvate carboxylase is involved in gluconeogenesis, lipogenesis, insulin secretion and synthesis of the neurotransmitter glutamate. Mutations in this gene have been associated with pyruvate carboxylase deficiency. Alternatively spliced transcript variants with different 5' UTRs, but encoding the same protein, have been found for this gene. [provided by RefSeq, Jul 2008]. * **<color #00a2e8>UniProt Summary</color>**: Pyruvate carboxylase catalyzes a 2-step reaction, involving the ATP-dependent carboxylation of the covalently attached biotin in the first step and the transfer of the carboxyl group to pyruvate in the second. Catalyzes in a tissue specific manner, the initial reactions of glucose (liver, kidney) and lipid (adipose tissue, liver, brain) synthesis from pyruvate. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |Biotin carb C| |Dala Dala lig C| |PYC OADA| |Biotin lipoyl| |CPSase L D2| |HMGL-like| |CPSase L chain| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |pyruvate carboxylase activity| |viral genome packaging| |viral RNA genome packaging| |positive regulation by host of viral release from host cell| |biotin binding| |modulation by host of viral release from host cell| |biotin metabolic process| |positive regulation of viral release from host cell| |positive regulation by host of viral process| |modulation by host of viral process| |regulation of viral release from host cell| |virion assembly| |gluconeogenesis| |hexose biosynthetic process| |monosaccharide biosynthetic process| |positive regulation of viral life cycle| |pyruvate metabolic process| |modification by host of symbiont morphology or physiology| |interaction with symbiont| |water-soluble vitamin metabolic process| |positive regulation of viral process| |modification of morphology or physiology of other organism involved in symbiotic interaction| |glucose metabolic process| |vitamin metabolic process| |carbohydrate biosynthetic process| |regulation of viral life cycle| |modification of morphology or physiology of other organism| |hexose metabolic process| |monosaccharide metabolic process| |viral life cycle| |regulation of viral process| |regulation of symbiosis, encompassing mutualism through parasitism| |coenzyme metabolic process| |sulfur compound metabolic process| |mitochondrial matrix| |cofactor metabolic process| |carbohydrate metabolic process| |drug metabolic process| |positive regulation of multi-organism process| |monocarboxylic acid metabolic process| |positive regulation of locomotion| |small molecule biosynthetic process| |viral process| |regulation of multi-organism process| |symbiotic process| |cellular amide metabolic process| |interspecies interaction between organisms| |carboxylic acid metabolic process| |regulation of locomotion| |oxoacid metabolic process| |organic acid metabolic process| |identical protein binding| |lipid metabolic process| |mitochondrion| |ATP binding| |negative regulation of gene expression| |small molecule metabolic process| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |Mubritinib 0.2μM R01 exp48|-3.02| |Nifuroxazide 5μM R03 exp102|-2.36| |Benzoate 10000μM R08 exp455|-1.91| |QNZ 0.01μM R08 exp439|-1.82| |Chloroquine 32μM R08 exp470|-1.75| |∆-9-Tetrahydrocannabinol 30μM R08 exp445|-1.72| |Pyridostatin 4μM R05 exp240|1.82| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 18/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|1/33| |central nervous system|1/56| |cervix|1/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|1/16| |kidney|1/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|1/24| |peripheral nervous system|1/16| |plasma cell|0/15| |prostate|1/1| |skin|1/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|3/22| |urinary tract|1/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 11202 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 5.58 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='PC Expression in NALM6 Cells: 5.58'> </modal> Last modified: 2026/01/07 22:36by 127.0.0.1
