The protein encoded by this gene is an integral membrane K-Cl cotransporter that can function in either a net efflux or influx pathway, depending on the chemical concentration gradients of potassium and chloride. The encoded protein can act as a homomultimer, or as a heteromultimer with other K-Cl cotransporters, to maintain chloride homeostasis in neurons. Alternative splicing results in two transcript variants encoding different isoforms. [provided by RefSeq, Sep 2008]. * **<color #00a2e8>UniProt Summary</color>**: Mediates electroneutral potassium-chloride cotransport in mature neurons and is required for neuronal Cl(-) homeostasis. As major extruder of intracellular chloride, it establishes the low neuronal Cl(-) levels required for chloride influx after binding of GABA-A and glycine to their receptors, with subsequent hyperpolarization and neuronal inhibition (By similarity). Involved in the regulation of dendritic spine formation and maturation (PubMed:24668262). {ECO:0000250|UniProtKB:Q63633, ECO:0000269|PubMed:24668262}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |AA permease| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |thermosensory behavior| |cellular chloride ion homeostasis| |potassium:chloride symporter activity| |hypotonic response| |cellular anion homeostasis| |cellular monovalent inorganic anion homeostasis| |chloride ion homeostasis| |chloride transmembrane transporter activity| |monovalent inorganic anion homeostasis| |cell volume homeostasis| |dendritic spine development| |potassium ion homeostasis| |potassium ion import across plasma membrane| |anion homeostasis| |inorganic cation import across plasma membrane| |inorganic ion import across plasma membrane| |response to osmotic stress| |multicellular organism growth| |chloride transmembrane transport| |import across plasma membrane| |chloride transport| |dendrite development| |inorganic anion transmembrane transport| |monovalent inorganic cation homeostasis| |learning| |inorganic anion transport| |potassium ion transmembrane transport| |potassium ion transport| |response to temperature stimulus| |regulation of cell size| |learning or memory| |anion transmembrane transport| |cognition| |regulation of cellular component size| |developmental growth| |growth| |monovalent inorganic cation transport| |chemical synaptic transmission| |anterograde trans-synaptic signaling| |trans-synaptic signaling| |synaptic signaling| |regulation of anatomical structure size| |inorganic cation transmembrane transport| |behavior| |anion transport| |cation transmembrane transport| |metal ion homeostasis| |metal ion transport| |cellular ion homeostasis| |inorganic ion transmembrane transport| |neuron projection development| |import into cell| |cation homeostasis| |inorganic ion homeostasis| |cellular chemical homeostasis| |ion homeostasis| |neuron development| |cation transport| |cellular homeostasis| |ion transmembrane transport| |neuron differentiation| |response to drug| |chemical homeostasis| |cell-cell signaling| |plasma membrane bounded cell projection organization| |cell projection organization| |response to abiotic stimulus| |transmembrane transport| |ion transport| |nervous system process| |integral component of plasma membrane| |generation of neurons| |neurogenesis| |homeostatic process| |cell development| |system process| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='default' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |LJH685 50μM R08 exp433|1.74| |YM155 0.0002μM R01 exp61|1.77| |BRD2 inhibitor II 20μM R07 exp341|1.81| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> No correlation found to any other genes in chemogenomics. </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 9443 * **<color #00a2e8>Expression level (log2 read counts)</color>**: 1.99 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='SLC12A5 Expression in NALM6 Cells: 1.99'> </modal> Last modified: 2026/01/07 22:37by 127.0.0.1
