Probably functions as a survival factor. Conjugation ability activated by UBA6. Promotes the expression of the proteasome subunit beta type-9 (PSMB9/LMP2). Regulates TNF-alpha-induced and LPS-mediated activation of the central mediator of innate immunity NF-kappa-B by promoting TNF-alpha-mediated proteasomal degradation of ubiquitinated-I-kappa-B-alpha. Required for TNF-alpha-induced p65 nuclear translocation in renal tubular epithelial cells (RTECs). May be involved in dendritic cell (DC) maturation, the process by which immature dendritic cells differentiate into fully competent antigen-presenting cells that initiate T-cell responses. Mediates mitotic non-disjunction and chromosome instability, in long-term in vitro culture and cancers, by abbreviating mitotic phase and impairing the kinetochore localization of MAD2L1 during the prometaphase stage of the cell cycle. May be involved in the formation of aggresomes when proteasome is saturated or impaired. Mediates apoptosis in a caspase-dependent manner, especially in renal epithelium and tubular cells during renal diseases such as polycystic kidney disease and Human immunodeficiency virus (HIV)- associated nephropathy (HIVAN). {ECO:0000269|PubMed:15831455, ECO:0000269|PubMed:16495226, ECO:0000269|PubMed:16495380, ECO:0000269|PubMed:17889673, ECO:0000269|PubMed:18574467, ECO:0000269|PubMed:19028597, ECO:0000269|PubMed:19033385, ECO:0000269|PubMed:19166848, ECO:0000269|PubMed:19726511, ECO:0000269|PubMed:19959714}. <button type='primary' size='sm' modal='Pfam_Domains'>Pfam Domains</button> <button type='primary' size='sm' modal='GO_terms'>GO Terms</button> <modal id='Pfam_Domains' size='lg' title='Pfam Domains'> |ubiquitin| </modal> <modal id='GO_terms' size='lg' title='GO Terms'> |aggresome assembly| |inclusion body assembly| |proteasome binding| |myeloid dendritic cell differentiation| |myeloid dendritic cell activation| |dendritic cell differentiation| |aggresome| |myeloid leukocyte differentiation| |fibrillar center| |response to interferon-gamma| |positive regulation of I-kappaB kinase/NF-kappaB signaling| |myeloid cell differentiation| |regulation of I-kappaB kinase/NF-kappaB signaling| |response to tumor necrosis factor| |leukocyte differentiation| |post-translational protein modification| |regulation of mitotic cell cycle phase transition| |regulation of cell cycle phase transition| |ubiquitin-dependent protein catabolic process| |modification-dependent protein catabolic process| |modification-dependent macromolecule catabolic process| |hemopoiesis| |myeloid leukocyte activation| |proteolysis involved in cellular protein catabolic process| |hematopoietic or lymphoid organ development| |cellular protein catabolic process| |regulation of mitotic cell cycle| |positive regulation of apoptotic process| |immune system development| |positive regulation of programmed cell death| |protein catabolic process| |protein ubiquitination| |positive regulation of cell death| |regulation of cell cycle process| |innate immune response| |protein modification by small protein conjugation| |cellular macromolecule catabolic process| |leukocyte activation| |defense response to other organism| |protein modification by small protein conjugation or removal| |positive regulation of intracellular signal transduction| |macromolecule catabolic process| |organonitrogen compound catabolic process| |cell activation| |response to cytokine| |regulation of cell cycle| |proteolysis| |response to other organism| |response to external biotic stimulus| |response to biotic stimulus| |defense response| |regulation of apoptotic process| |regulation of programmed cell death| |positive regulation of signal transduction| |regulation of cell death| |organic substance catabolic process| |cellular catabolic process| |positive regulation of cell communication| |positive regulation of signaling| |regulation of intracellular signal transduction| |immune response| </modal> \\ === CRISPR Data === <button type='primary' size='small' modal='Compound_Hit'>Compound Hit</button> <button type='primary' size='small' modal='Most_Correlated_Genes'>Most Correlated Genes in Chemogenomics</button> <button type='primary' size='small' modal='Essential_Avana'>Tissues where Essential in the Avana Dataset (DepMap 20Q1)</button> <modal id='Compound_Hit' size='lg' title='Compound Hit'> ^Screen^Score^ |CCT251545 20μM R07 exp326|-1.78| |Beta-Estradiol 0.55μM R07 exp425|-1.75| |Losmapimod 1μM R07 exp376|-1.74| </modal> <modal id='Most_Correlated_Genes' size='lg' title='Most Correlated Genes in Chemogenomics'> ^Gene^Correlation^ |RRM1|0.493| </modal> <modal id='Essential_Avana' size='lg' title='Tissues where Essential in the Avana Dataset (DepMap 20Q1)'> Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| </modal> == Essentiality in NALM6 == * **<color #00a2e8>Essentiality Rank</color>**: 18801 * **<color #00a2e8>Expression level (log2 read counts)</color>**: -7.68 <button type='primary' size='small' modal='Dist_expr'>Expression Distribution</button> <modal id='Dist_expr' size='lg' title='UBD Expression in NALM6 Cells: -7.68'> </modal> Last modified: 2026/01/07 22:37by 127.0.0.1
